What could be wrong if my calves are hurting while running?

If you have just started running or if you are running for the first time after a break you will probably have some pain or discomfort in your calves. If it continues after that or is unbearable you should talk to a doctor.

Many of the pains I experienced when I started running were due to wearing the wrong shoes. My feet and gait were analyzed at my local specialty running store by a person, trained in podiatry. He recommended more suitable shoes, which did alleviate my pains, and I've stuck with that model since then.

You might have pulled a muscle, or might not have stretched them properly before you started to run (or wearing the wrong type of shoe).

You may be going too fast. New runners frequently start out going too fast and find that they are heaving for breath or have sore calves when running, even if they're normally in good shape. To prevent calf pain, make sure you take rest days and stretch before and after every workout.

If your calves are hurting while you are running, there could be a problem that you are over training that muscle group, and need to rest.
