Practice your lines multiple in front of a mirror, and record it if possible (use your cellphone if you don't have a recorder) to listen to how you sound. The more you know the lines the more confident you will be, and thus will do a better job. Remember to breathe.

Know the character you want to play. If you want to play Crystal, Ronnette, or Chiffon, you need to emphasize your ability to sing harmony parts. For the Dentist, you need to sing a piece in a low range. For Seymour, a high range. If you want to play Audrey, practice an accent and sing with it.
